A Robbie Keane goal in the first half set up a fourth round FA Cup tie against Manchester United.
Keane finished from close range after Younes Kaboul had headed a Jermaine Jenas corner against the bar after 15 minutes.
Reading's best chance came when substitute Stephen Hunt hit the bar with a fierce shot.
Steed Malbranque came closest to added a second for Spurs, when his shot was tipped onto the bar by Adam Federici.
Spurs reward is a trip to Old Trafford in the next round.
